RESUMO

CircRNAs are a class of non-coding RNAs able to regulate gene expression at multiple levels. Their involvement in physiological processes, as well as their altered regulation in different human diseases, both tumoral and non-tumoral, is well documented. However, little is known about their involvement in female reproduction. This study aims to identify circRNAs potentially involved in reproductive women's health. Candidate circRNAs expressed in ovary and sponging miRNAs, already known to be expressed in the ovary, were selected by a computational approach. Using real time PCR, we verified their expression and identified circPUM1 as the most interesting candidate circRNA for further analyses. We assessed the expression of circPUM1 and its linear counterpart in all the follicle compartments and, using a computational and experimental approach, identified circPUM1 direct and indirect targets, miRNAs and mRNAs, respectively, in cumulus cells. We found that both circPUM1 and its mRNA host gene are co-expressed in all the follicle compartments and proposed circPUM1 as a potential regulator of PTEN, finding a strong positive correlation between circPUM1 and PTEN mRNA. These results suggest a possible regulation of PTEN by circPUM1 in cumulus cells and point out the important role of circRNA inside the pathways related to follicle growth and oocyte maturation.

In the context of the circular economy, the development of innovative and low-carbon concrete that incorporates different kinds of waste materials is gaining attention among the research community, regulatory agencies, and policymakers. These materials can be incorporated into concrete mixtures as aggregates or as fillers for improvement of product properties. This study aims to identify reliable designs for biochar-augmented cementitious products and general applications through technical, environmental, and economic assessments. The outcomes demonstrate that 5 wt% biochar addition could enhance the compressive strength of the final products. Using biochar, together with other recycled materials, can enormously reduce the environmental impacts, especially for global warming, enabling biochar-augmented cementitious products and general application as carbon-negative resources. The highest GWP reduction reached -720 kg CO2/tonne, equal to a 200% saving. A high quantity of biochar could be included in several specific applications (up to 60 wt%). The economic assessment highlights that the proposed designs are cost-effective and carbon tax can be significantly reduced. Carbon credits can also be earned for some carbon-negative designs. These findings can serve to mitigate GHG emissions and provide decision-makers with a reliable and holistic framework towards the goal of carbon neutrality.

Lanthanide-doped nanoparticles, featuring sharp emission peaks with narrow bandwidth, exhibit high downconversion luminescence intensity, making them highly valuable in the fields of bioimaging and drug delivery. High-crystallinity Y2O3 nanoparticles (NPs) doped with Er3+ ions were functionalized by using a pegylation procedure to confer water solubility and biocompatibility. The NPs were thoroughly characterized using transmission electron microscopy (TEM), inductively coupled plasma mass spectrometry (ICP-MS), and photoluminescence measurements. The pegylated nanoparticles were studied both from a toxicological perspective and to demonstrate their internalization within HCT-116 cancer cells. Cell viability tests allowed for the identification of the "optimal" concentration, which yields a detectable fluorescence signal without being toxic to the cells. The internalization process was investigated using a combined approach involving confocal microscopy and ICP-MS. The obtained data clearly indicate the efficient internalization of NPs into the cells with emission intensity showing a strong correlation with the concentrations of nanoparticles delivered to the cells. Overall, this research contributes significantly to the fields of nanotechnology and biomedical research, with noteworthy implications for imaging and drug delivery applications.

The partial degradation of cellulose-based bioplastics in industrial treatment of organic fraction of Municipal Solid Waste (MSW) opened to the investigation of further disposal routes for bioplastics in the waste management system. For this purpose, the environmental footprint of three MSW management scenarios differing only for the bioplastics final destination (organic, plastic or mixed waste streams) was assessed through a Life Cycle Assessment (LCA) approach. Results revealed how the treatment of bioplastics with organic waste achieved the worst environmental performance (5.8 kg CO2 eq/FU) for most impact categories. On the other hand, treatment with plastics and mixed waste achieved negative impact values (that mean avoided GHG emissions) of -9.8 and -7.7 kg CO2 eq/FU respectively, showing comparable benefits from these scenarios. The key reason was the lower quality of compost obtained from the organic treatment route, which reduced the environmental credits achieved by the energy recovery during anaerobic digestion.

The main aim of this study was to perform a combined life cycle assessment and life cycle costing of the most used wine packaging systems in Italy. Packaging plays a key role in food and beverage production and supply chain, but the increasing volume of packaging used causes many environmental concerns. The wine sector is no exception, especially in Italy that is the largest producer in the world. Eight packaging alternatives were compared in environmental and economic terms assuming a functional unit of three litres and a cradle to grave approach: one aseptic carton system, two bag-in-box systems, two PET bottle systems, and three glass bottle systems. The results were analysed with an eco-efficiency portfolio approach. There was consistency between the LCA and LCC results for most of the packaging systems. From both the environmental and economical point of views, aseptic cartons and bag-in-box systems were the most sustainable alternatives, while the glass systems had the worst global performances due to the high weight and consequent huge energy consumption during bottle production. The size of the containers was the key factor that most affected the results both in environmental and economic terms.

Bacteria are believed to play an important role in intestinal tumorigenesis and contribute to both gut luminal and circulating metabolites. Celecoxib, a selective cyclooxygenase-2 inhibitor, alters gut bacteria and metabolites in association with suppressing the development of intestinal polyps in mice. The current study sought to evaluate whether celecoxib exerts its chemopreventive effects, in part, through intestinal bacteria and metabolomic alterations. Using ApcMin/+ mice, we demonstrated that treatment with broad-spectrum antibiotics (ABx) reduced abundance of gut bacteria and attenuated the ability of celecoxib to suppress intestinal tumorigenesis. Use of ABx also impaired celecoxib's ability to shift microbial populations and gut luminal and circulating metabolites. Treatment with ABx alone markedly reduced tumor number and size in ApcMin/+ mice, in conjunction with profoundly altering the metabolite profiles of the intestinal lumen and blood. Many of the metabolite changes in the gut and circulation overlapped and included shifts in microbially derived metabolites. To complement these findings in mice, we evaluated the effects of ABx on circulating metabolites in patients with colon cancer. This showed that ABx treatment led to a shift in blood metabolites, including several that were of bacterial origin. Importantly, changes in metabolites in patients given ABx overlapped with alterations found in mice that also received ABx. Taken together, these findings suggest a potential role for bacterial metabolites in mediating both the chemopreventive effects of celecoxib and intestinal tumor growth. PREVENTION RELEVANCE: This study demonstrates novel mechanisms by which chemopreventive agents exert their effects and gut microbiota impact intestinal tumor development. These findings have the potential to lead to improved cancer prevention strategies by modulating microbes and their metabolites.

Advanced maternal age impairs reproductive performance, influencing the quantity and the quality of oocytes. Mitochondria dysfunction seems to play a decisive role in conditioning the quality of the female gamete. Different in vitro and in vivo studies, demonstrated the antioxidant and anti-inflammatory activities of Resveratrol and its ability to improve mitochondria function even if the exact mechanism of action has not yet been demonstrated in human oocytes. In this paper, by retrospective analysis, we evaluated follicular fluid (FF) miRNome modification in aged women with a poor ovarian reserve receiving a resveratrol-based supplement the three months before the in vitro Fertilization (IVF) cycle. We found 13 differentially expressed microRNAs (miRNAs) in women treated with resveratrol and specifically miR-125b-5p, miR-132-3p, miR-19a-3p, miR-30a-5p and miR-660-5p, regulating mitochondrial proteins, are able to control metabolism and mitochondrial biogenesis. MiRNA expression differences, observed after resveratrol treatment in FF from women with a poor prognosis for IVF, demonstrated that resveratrol may act on mitomiRNAs to improve follicular microenvironment by transcriptomic and proteomic modifications in granulosa cells.

PURPOSE: Long non-coding RNAs (lncRNAs) control gene expression at multiple levels. By interacting with microRNAs (miRNAs), they regulate their mRNA targets creating dynamic regulatory networks involved in different cellular processes. Their role in follicle development and oocyte maturation has recently emerged. lncRNA deregulation has been found associated with different pathological conditions. In this study, we identified differentially expressed lncRNAs in cumulus cells (CCs) isolated from MII oocytes of advanced maternal age women and proposed ceRNA-networks involved in signaling pathways crucial in ovarian folliculogenesis and female germ cell maturation. METHODS: We performed a high-throughput analysis of the expression profile of 68 lncRNAs from CCs of aged and young women by using NanoString technology. By miRNet, TarPmiR, miRTarBase, OKdb, and KEGG we predicted some ceRNA-networks involving the differentially expressed (DE) lncRNAs, miRNA interactors, and their mRNA target genes. RESULTS: We identified 28 lncRNAs down-regulated in CC samples from aged women. The analysis revealed that the miRNAs binding 11 of the DE lncRNAs and their mRNA targets are included in ceRNA-networks involved in the regulation of the PI3K-Akt, FOXO, and p53 signaling pathways. CONCLUSION: We proposed that the lncRNA down-regulation in CCs from aged women could influence the expression of genes encoding proteins deregulated in reproductive aging. A better understanding of the interplay of lncRNA-miRNA-mRNA networks in human CCs could increase our knowledge about the mechanisms of regulation of gene expression involved in aging, lead to the development of novel therapeutics, and improve reproductive outcomes in aged women.

In the last few years, microRNA-mediated regulation has been shown to be important in viral infections. In fact, viral microRNAs can alter cell physiology and act on the immune system; moreover, cellular microRNAs can regulate the virus cycle, influencing positively or negatively viral replication. Accordingly, microRNAs can represent diagnostic and prognostic biomarkers of infectious processes and a promising approach for designing targeted therapies. In the past 18 months, the COVID-19 infection from SARS-CoV-2 has engaged many researchers in the search for diagnostic and prognostic markers and the development of therapies. Although some research suggests that the SARS-CoV-2 genome can produce microRNAs and that host microRNAs may be involved in the cellular response to the virus, to date, not enough evidence has been provided. In this paper, using a focused bioinformatic approach exploring the SARS-CoV-2 genome, we propose that SARS-CoV-2 is able to produce microRNAs sharing a strong sequence homology with the human ones and also that human microRNAs may target viral RNA regulating the virus life cycle inside human cells. Interestingly, all viral miRNA sequences and some human miRNA target sites are conserved in more recent SARS-CoV-2 variants of concern (VOCs). Even if experimental evidence will be needed, in silico analysis represents a valuable source of information useful to understand the sophisticated molecular mechanisms of disease and to sustain biomedical applications.

Despite providing important ecological functions, seagrass accumulation causes environmental and economic issues, including eutrophication and tourism reduction. Nowadays, seagrass wrack is commonly removed from the beaches and landfilled, which is considered the least desirable practice according to the European Union (EU) Waste Framework Directive. In this study, different management strategies for seagrass valorisation, including anaerobic digestion (AD), composting and ecological restoration, were considered using a life cycle assessment (LCA) perspective. The aim of the work was to evaluate more ecological and economic alternatives to landfill and to provide a robust evaluation method for public and private companies. An economic assessment was subsequently conducted, considering both direct and indirect impacts with a life cycle costing (LCC) approach. A selected beach located in the Northeast Mediterranean Sea was considered as a relevant case-study. The environmental impacts of the seagrass management scenarios were evaluated with the method ReCiPe 2016H, using both midpoint and endpoint levels. LCA results showed that ecological restoration and AD were the best alternatives in terms of environmental performances because of biogas production used as a renewable energy source. The impacts of the alternative management strategies were significantly lower than the current landfill strategy, -70% considering the categories of human health, ecosystems and resources, and -95% considering global warming potential category. The LCC analysis proved that composting was the best alternative (NPV > 1.27 M€), due to lower operating costs and higher fertilizer value. The obtained results can help beach management companies and public administrations to select the best operational strategies to reduce the environmental and economic impact of seagrass collection and treatment.

BACKGROUND: Despite availability of clinical guidelines, underdiagnosis, undertreatment, and poor adherence are still significant concerns in allergic rhinitis (AR) therapeutic management. We investigated clinical practice patterns and prescribing behavior of Italian healthcare professionals (HCPs) specialized in AR. METHODS: One-hundred allergologists, 100 ear, nose and throat (ENT) specialists, and 150 general practitioners (GPs) were recruited. The survey assessed: socio-demographic, work experience, monthly caseload, prescription drivers. Next, HCPs were invited to retrospectively recover patients' clinical data to investigate: AR clinical characteristics, therapy management, prescription patterns, patient adherence. Descriptive statistics, Chi square, One-Way analysis of variance, and Two-Way Analysis of Variance were performed. RESULTS: Allergologists visited more AR patients (31% of monthly caseload) than ENTs (21%, p < 0.001), while GPs' caseload was the lowest (6%). Clinical information of 2823 patients were retrieved of whom 1906 (67.5%) suffered from moderate/severe AR (discomfort score: 7.7 ± 1.3) and 917 (32.4%) from mild AR (5.7 ± 1.9). About one-third of mild patients had a discomfort score ≥ 7. Main prescription drivers were "effective on all symptoms" (54.3% patients) and "quick symptom relief" (47.8%), whereas minor drivers were "affordable price" (13.4%) and "refundable" (8.7%). The most prescribed drugs were antihistamines and intranasal corticosteroids (79% and 55% prescriptions), followed by fixed-dose-combination of intranasal azelastine/fluticasone (19%). Polytherapy was the most common treatment strategy (59.6%). HCPs' believe that the majority of the patients was adherent to treatment (88% with score > 7). CONCLUSIONS: This survey describes the therapeutic approach adopted by Italian physicians to cope with AR and shows that HCPs underestimated AR severity and had a non-realistic perception of patients' adherence. These findings suggest that further efforts are required to improve AR clinical management in Italy.

This study presents an evaluation of the environmental performance of an ice cream cup made of polyethylene (PE)/paper laminate using a life cycle assessment approach 'from cradle to grave'. Two opposite alternative disposal scenarios, as well as their intermediate combinations, were considered: 100% incineration and 100% landfilling. The environmental impacts were calculated using the EPD 2013 evaluation method since the study was developed in an Environmental Product Declaration perspective as well as the method ReCiPe 2008 H at the endpoint level. PE/paper laminate production was the most impactful process since it provided the highest contribution to total impacts in four of six impact categories considered. Ice cream cup production was the second impactful process. The 100% incineration scenario provided negligible contribution to life cycle total impact for all impact categories; while considering the landfilling scenario, the percentage contributions to the total impact provided by the end-of-life phase increased considerably, until to be comparable to the contributions provided by the production processes of the PE/paper laminate and the ice cream cup. The obtained results highlighted that different disposal scenarios can affect significantly the conclusions of a study. At the endpoint level, incineration was more environmentally sound than landfilling for all the ReCiPe damage categories.

The main aim of this study was to perform a Life cycle assessment study as well as an economic evaluation of the recovery of recyclable materials in a municipal solid waste management system. If citizens separate erroneously waste fractions, they produce both environmental and economic damages. The environmental and economic evaluation was performed for the case study of Nola (34.349 inhabitants) in Southern Italy, with a kerbside system that assured a source separation of 62% in 2014. The economic analysis provided a quantification of the economic benefits obtainable for the population in function of the achievable percentage of source separation. The comparison among the environmental performance of four considered scenarios showed that the higher the level of source separation was, the lower the overall impacts were. This occurred because, even if the impacts of the waste collection and transport increased, they were overcome by the avoided impacts of the recycling processes. Increasing the source separation by 1% could avoid the emission of 5 kg CO2 eq. and 5 g PM10 for each single citizen. The economic and environmental indicators defined in this study provide simple and effective information useful for a wide-ranging audience in a behavioural change programme perspective.

This paper presents the results of a survey regarding customer satisfaction as well as other aspects relating to the frequency and motivations of citizens going to a separate collection centre (SCC) for recyclables in a Southern Italian town. The study was carried out in 2015 to verify whether there were significant changes in the behaviours and opinions of the SCC's users, who had been interviewed in 2013, in the light of the change of the local administration. The majority of the respondents (60.4%) coupled going to the SCC with other tasks, with 58.6% of them going to a supermarket. Therefore, future SCCs should localized near shopping centres (it would be preferable to have small and numerous SCCs). Chi-square and Fisher's exact tests showed how SCC's users of the town under study behaved as a community. The percentage of respondents that declared to 'agree' or 'strongly agree' with the idea that only putrescibles and residue could be collected at their home, with all the other materials being collected at the SCC, increased from 56.6%, in 2013, up to 59.0%, in 2015. The percentage of respondents that declared going to the SCC due to environmental motivations was unanimous. Moreover, the saving of money for the community became the second preferred option, with a significant improvement (from 65.6%, in 2013, up to 93.2%, in 2015). Therefore, the community's interests over the individual's were privileged. The SCC's users confirmed their attention to environmental issues even though the local administration changed. It is probable that this could be the result of maintaining high standard of service as well as a good job in terms of communication, continuous involvment of citizens and the adoption of good environmental practices.
